EARLY ASSESSMENT OF CHANGES IN MITOCHONDRIAL HEALTH

Our Ready-2-Go Mitochondrial Health Assay Service assesses changes in mitochondrial membrane potential that result from treatment with your drug candidates. Mitochondrial dysfunction is frequently an early indicator of apoptosis and cellular toxicity, and our assay measures mitochondrial health with a fluorescent dye that accumulates within mitochondria proportionately to mitochondrial membrane potential. Combining this with our Ready-2-Go Cell Health Assay Service provides you with a general toxicity framework of your drug candidates that can save you time and money in your drug-development campaign.

Any and all! Mitochondrial membrane potential is indispensable to supporting ATP synthesis, ROS generation, and calcium uptake/storage. Membrane depolarization is an essential indicator of mitochondrial dysfunction, which is increasingly implicated in drug-candidate attrition and post-market withdrawals in myriad disease areas.

ASSAY OUTLINE

Cells are seeded and cultured in 384-well plates. They are treated with known mitochondrial stressors (rotenone, oligomycin A, and FCCP) along with your test articles for 4 and 24 hours, at which points the cells are fixed and stained with mitochondrial indicator dyes. Cells are then imaged on a high-content microscope, and images are analyzed to provide you quantitative assessments of your compounds’ effects on mitochondrial function.
