Now that we have two ROIs, higher intensity punctate subregions or “spots” can be segmented in a method similar to that used in step 2 to detect the nucleus.
Spots can be quantified in terms of count, area (both average and total per cell), average and total pixel intensity, shape texture and in more advance assays even colocalization with other protein signals.
